Do you need a good IP antibody to your protein of interest?
Perform an IP on a GFP-tagged fusion protein.
A GFP SILAC co-IP is almost exactly the same as a SILAC co-IP, except that the target protein is expressed from an artificial construct as a fusion with GFP.
Care must be taken that fusion expression levels are as close to physiological levels as possible, and that fusion subcellular localisation is similar to that of wt protein. It is also better to perform the experiment using both N- and C-terminal fusions.
NB: While this service describes performing IPs on GFP-tagged fusion proteins, the same type of proteomics analysis can be performed with little modification on proteins tagged with other tags, as long as these can be used to IP the protein.
